I looked and the only posts that refer to this Error -5018 are a few years old. I wondered if anyone has had this error with InstallShield 2009?
This error is occuring in InstallShield 2009 SP2, running in an Administrator account. Here is the pertinant portion of the log file. I am not including the first part of the log where the two warnings occur. They have to do with prerequisites and do not pertain to this particular problem.

There's nothing obvious in the code which suggests to me why this happens. Are you using any unusual source control or antivirus solutions which might render the building setup.exe read only? You could also try to watch this in Process Monitor to see if there are any indications why the write to the setup.ini file fails.
